When the channel is said to be pinched –off?

If voltage between drain and source Vds becomes sufficiently large that Vgd < Vt , the channel is no longer inverted near the drain and channel becomes pinched off.

Where Vg = gate-to-drain voltage, Vt= Threshold voltage.
